DePick: Wnt Inhibitors Assay

The predicted targets for the "Wnt inhibitors assay" project were: MAOB, ALDH2, APP, MCHR1, ABL1, MTOR, HMGCR, CYP19A1, and CYP1B1. The canonical Wnt / beta-catenin signaling pathway is a highly conserved and essential signaling pathway involved in organ growth and maintenance, which has also been implicated in several pathologies like e.g. cancer. The displayed graph shows protein targets that regulate the main player of the wnt signaling pathway beta-catenin (CTNNB1). The proteins influence CTNNB1 either directly, or indirectly via the alteration of metabolite levels. A direct interactor is e.g. ABL1. It interacts with and stabilizes hereby CTNNB1. Indirect interactors are e.g. HMGCR, ALDH2, and MAOB. HMGCR is a central enzyme of cholesterol biosynthesis pathway and has therefore an regulatory effect on CTNNB1 via cholesterol. ALDH2 and MAOB have been reported to regulate the serotonin level. Serotonin is known to inhibit the central Wnt signalling inhibitor GSK3B.
